Clinical Significance Decreased antithrombin III activity, protein C activity, free protein S, and the presence of a factor V (Leiden) and/or factor II 20210G>A mutation each contribute to an increased risk of hereditary thrombophilia. Approximately 10% of factor V (Leiden) heterozygotes also carry the prothrombin 20210G>A mutation. Likewise, protein C and/or S deficiency have been reported with factor V (Leiden) mutation. Since venous thrombosis is a multifactorial disorder, presence of a second risk factor further increases the thrombotic risk.
